چکیده

The mechanism of the fluorescence quenching of aqueous thionine by various metal ions (Ce3+_, Fe2+, Mn2+, Mg2+, Cs+ and Na+) was studied at pn = 2.5. The following Stern-Volmer constants (K s v) were determined: XSv(Fe2+) == 1.13 dm3 mol -1 , Ksw(Mn2+) = 0.24 dm3 mol"1, Ksv(Ce3+) = 0.24 dm3 mol -1, i£sv(Cs~) — 0.03 dm3 mol -1 . At concentrations below 1 mol.dm -3 Na2S04 and MgS04 are very weak quenchers (Ksv(Na+ or Mg2+) = 0.007), but the influence on the fluorescence yield increases considerably at higher cation concentrations. This was attributed to the formation of ground state complexes between thionine and SO4 ions. The salt concentration affects considerably the quenching rate of Fe2+ ions, but much less so that of the other cations. Fluorescence quenching mechanisms are discussed.

Photo-induced inactivation of viruses: adsorption of methylene blue, thionine, and thiopyronine on Qbeta bacteriophage.

The adsorption of cationic organic dyes (methylene blue, thionine, and thiopyronine) on Qbeta bacteriophage was studied by UV-visible and fluorescence spectroscopy. The dyes have shown a strong affinity to the virus and some have been used as sensitizers for photo-induced inactivation of virus. In the methylene blue concentration range of 0.1-5 microM and at high ratios of dye to virus (greater...

Fluorescence quenching of carbon nitride nanosheet through its interaction with DNA for versatile fluorescence sensing.

This work investigates the interaction of carbon nitride nanosheet (CNNS), a recently developed two-dimensional nanomaterial, with DNA and its fluorescence quenching mechanism on fluorophore labeled single-stranded DNA probes. The static quenching through the photoinduced electron transfer (PET) from the excited fluorophore to the conductive band of CNNS is identified. Utilizing the affinity ch...

Tryptophan fluorescence quenching as a monitor for the protein conformation changes occurring during the photocycle of bacteriorhodopsin under different perturbations.

The rates of the quenching and recovery of tryptophan fluorescence are determined in the microsecond-millisecond time scale during the photocycle of bacteriorhodopsin under different perturbations. The kinetics suggest the presence of two quenching processes, a rapid one (on the time scale of photocycle intermediate L550 formation or faster) and a slow one (slightly slower than the slow compone...

A novel fluorescence "on-off-on" chemosensor for Hg(2+)via a water-assistant blocking heavy atom effect.

Upper rim pyrene-functionalized hexahomotrioxacalix[3]arene L was synthesized via Click chemistry, and its fluorescence behaviors toward several common metal cations were investigated. L exhibited a significant fluorescence quenching response to Hg(2+) in CH3CN solution, which was unaffected by the coexistence of other competitive metal cations. Thus, L can be utilized as a highly selective and...

Intrinsic tryptophan fluorescence as a probe of metal and alpha-ketoglutarate binding to TfdA, a mononuclear non-heme iron dioxygenase.

2,4-Dichlorophenoxyacetic acid (2,4-D)/alpha-ketoglutarate (alphaKG) dioxygenase, TfdA, couples the oxidative decarboxylation of alphaKG to the oxidation of the herbicide 2,4-D using a mononuclear non-heme Fe(II) active site. The intrinsic tryptophan fluorescence associated with the four Trp residues in TfdA allows for the use of fluorescence spectroscopy to monitor the binding of iron and alph...
